Es werden stationäre und zeitaufgelöste Fluoreszenzmessungen ausgeführt. Ziel ist, aus der Fluoreszenzkinetik der Tryptophane Rückschlüsse auf Veränderungen in der Proteinstruktur als Folge von Induktor- bzw. DNA-Bindung zu ziehen. Als weitere Sonde kann auch die Fluoreszenz des Induktors (Tetrazykline als Komplex mit 2-wertigen Metallionen) herangezogen werden.
Anwendung von Hochdruck erlaubt zusätzlich partiell denaturierte Zustände zu erfassen. Publikationen:
